چکیده

تهیه و توسعه‌ی عامل‌های تشخیص عفونت‌ها در تشخیص گونه‌های مقاوم به درمان، به دلیل مرگ و میر ناشی از عفونت این نوع گونه‌ها از جمله باکتری قارچی سل ضرورت مهمی در مطالعه‌های بالینی تلقی می‌شود. در این مطالعه‌، شرایط مختلف محیطی برای نشان‌دارسازی سریع و کارآمد آنتی‌بیوتیک ریفامپیسین به وسیله‌ی تکنسیم-m99 برای استفاده‌ی نهایی از آن در تشخیص بالینی عفونت‌های سل بهینه‌سازی شد. خلوص رادیوشیمیایی به روش کروماتوگرافی سریع لایه‌ی نازک بر روی کاغذ واتمن شماره‌ی 1 در حلال متیل‌اتیل‌کتون و سالین نرمال کنترل شد. عامل‌های زمان، دما، مقدار یون قلع و pH محیط در فرایند نشان‌دارسازی مورد مطالعه قرار گرفتند؛ دمای اتاق (27 درجه‌ی سانتی‌گراد)، 7=pH، مقدار 20 میکروگرم قلع کلرید، شرایط بهینه برای نشان‌دارسازی یک میلی‌گرم ریفامپیسین با مقدار 2 میلی‌کوری تکنسیم-m99 به شکل پرتکنتات تازه دوشیده شده از مولد را فراهم نمود. تحت این شرایط خلوص رادیوشیمیایی کمپلکس نشان‌دار 85 تا %90 تعیین شد. کمپلکس نشان‌دار پایداری قابل توجهی در حضور سرم انسانی در 37 درجه و در محلول نهایی رادیودارو تا 6 ساعت از خود نشان داد.

Developing of new infection imaging agents is a mandate in the detection of resistant species in the clinic due to the mortality of various new strains of bacteria including mycobacterium tuberculosis. In this study, various conditions were optimized for the rapid and efficient labeling of rifampicin antibiotic labeled with Tc-99m for ultimate use in infection imaging. Radiochemical purities were checked by RTLC using methyl ethyl ketone, and normal saline on Whatman No.1 paper. The time, temperature, ligand concentration, stannous ion amount, pH were optimized in the radiolabeling process and the best conditions were room temperature, pH 7, 20 micrograms of stannous chloride for 1 mg of rifampicin solid and 20 mCi of freshly milked technetium-99m pertechnetate. The complex demonstrated satisfactory stability in the presence of human serum and final formulations for 6 hours.
