چکیده
در این کار تحقیقاتی، تولید ویتامین 12B نشاندار با استفاده از گونهای از اکتینومیست در یک محیط کشت جدید به روش تخمیر ناپیوسته مورد بررسی قرار گرفته است. پس از تولید و آزادسازی ویتامین 12Bاز سلولهای میکروبی، محلول حاوی ترکیبات کوبالامین روی ستونی از رزین XAD-4 تثبیت شده و ستون توسط محلولهای مختلف شسته شد. مقدار ویتامین 12B موجود در محلول خروجی از ستون، به روش کروماتوگرافی مایع با کارایی بالا اندازهگیری شد و جهت تأیید وجود ترکیبات کوبالامین در نمونهها، از روش کروماتوگرافی لایهی نازک استفاده گردید. نتایج نشان داد که میزان تولید ویتامین 12B توسط این میکروارگانیسم 1456 میکروگرم در لیتر است. اتانول و استون، به عنوان بهترین محلولهای شستشوی ستون برای جداسازی ویتامین 12B تعیین شدند. اندازهگیریهای کروماتوگرافی لایهی نازک نشان داد که مقادیر Rf سیانوکوبالامین و متیل کوبالامین تولید شده توسط باکتری با مقادیر استاندارد مطابقت میکنند. فعالیت ویژهی 12B نشاندار، پس از انجام مراحل واجذب آن با استفاده از عوامل مختلف استن، الکل خالص، الکل 25% و آب، به ترتیب، برابر با 8.55 ، 6.90 ، 0.79 و 5.75 بکرل در میلیلیتر اندازهگیری شد. در نهایت، محلولی از 12B نشاندار با فعالیت ویژهی 21.99 بکرل در میلیلیتر به دست آمد.

چکیده [English]
In this study, the production of labeled vitamin B12 with one strain of actinomycet in a novel synthetic medium by batch fermentation method was investigated. After releasing vitamin B12 from microbial cells, the solution containing cobalamin analogues was fixed on amberlite XAD-4 exchanger and eluted by various solutions. Vitamin B12 in the collected fractions were measured by HPLC method and also in order to confirm the existence of cobalamin compounds in the samples, TLC technique was used. In the optimal conditions, production of vitamin B12 by this microorganism was 1456µg/l. The best recoveries of vitamin B12 from the column were obtained when the column was eluted by ethanol followed by acetone. The Rf values for cyanocobalamin and methylcobalamin, produced by this microorganism were the same as the standard values of Rf. The desorption of labeled vitamine B12 was carried out by different eluants, including acetone, ethanol (absolute), ethanol 25% and deionized water. The activity of labeled vitamin B12 was 8.55, 6.90, 0.79, 5.75 Bq/ml, respectively. Therefore, the total specific activity of 21.99Bq/ml was obtained for labeled vitamin B12.
