تهیه و کنترل کیفی ترکیبات بلئومایسین نشاندار شده با رادیوایزوتوپ کبالت-57 برای مطالعه تومورهای سرطانی

چکیده
رادیوداروی [Co55] - بلئومایسین برای مطالعه تومورهای سرطانی کاربرد گسترده‌ای یافته است. با توجه به نیمه عمر کوتاه کبالت-55، برای انجام مطالعات اولیه و بررسیهای کامل در زمینه نشاندارسازی، استفاده ازرادیوایزوتوپ کبالت-57 به مراتب مناسبتر است. مطالعات جامع به منظور تعیین بهترین دما، اسیدیته، زمان و غلظت واکنشگرها برای نشاندارسازی با کبالت-57 انجام شد. نتایج اولیه نشان دادند که بلئومایسین (به مقدار 0.3 میلی‌گرم) در محیط اسیدی ضعیف تا خنثی در دمای اتاق و در حضور کلریدکبالت (0.4  میلی‌کوری)، در محیط آبی کمپلکس تشکیل می‌دهد. کنترلهای کیفی، از جمله کنترل رادیونوکلئیدی، رادیوشیمیایی و شیمیایی روی محصولات اولیه انجام گرفت که نشان‌دهنده امکان استفاده از این ترکیب در آزمونهای زیست‌شناختی بود. این نتایج در آینده نزدیک برای تهیه ترکیب [Co55] - بلئومایسین مورد استفاده قرار خواهند گرفت تا تصویربرداری و داده‌های زیست‌شناختی از آن تهیه شود.

Preparation and Quality Control of Radiolabelled Cobalt-57 Bleomycins for Oncological Studies

چکیده English

[55Co] bleomycin has been widely used in oncological studies. Due to relative short half life of cobalt-55, optimization and feasibility studies for determination of the best labeling conditions is preferably performed using cobalt-57. In this study, the optimization of complexation conditions for time, temperature and ligand concentration were performed. The best labeling yield was obtained at room temperature in pH of 4-7, using 0.4 mCi of 57CoCl2 with 0.3mg of BLM in 30 minutes. The final radiopharmaceutical solution underwent common quality control tests, validating our future studies for animal scanning studies using [55Co]bleomycin complex.
